Kisspeptin-10 is a reference agonist for studying the neuroendocrine control of the reproductive axis. This expanded guide covers the kisspeptin family, its receptor mechanism, and its role upstream of GnRH signaling.

Molecular identity & the kisspeptin family

Kisspeptins are products of the KISS1 gene, proteolytically processed into peptides including kisspeptin-54, -14, -13 and the C-terminal decapeptide kisspeptin-10. All share the C-terminal Arg-Phe-amide motif required for receptor activation, and kisspeptin-10 retains full activity at the receptor while being convenient to synthesize.

Mechanism of action (in vitro)

Kisspeptin-10 agonizes KISS1R (formerly GPR54), a class A GPCR coupling to Gαq/phospholipase-C. Activation generates IP3 and diacylglycerol, mobilizing intracellular calcium and activating ERK signaling in receptor-expressing cells.

Position in the neuroendocrine axis

On gonadotropin-releasing-hormone (GnRH) neurons, KISS1R activation is a principal upstream trigger of GnRH signaling, placing kisspeptin at the apex of hypothalamic–pituitary–gonadal (HPG) axis regulation. This makes kisspeptin-10 a central tool for studying reproductive neuroendocrine pathways in model systems.

Preclinical research landscape

Kisspeptin-10 is used in KISS1R binding, calcium-mobilization and ERK reporter assays and preclinical neuroendocrine models. Observations characterize receptor and pathway signaling and are not indicators of any human outcome.

Laboratory handling & reconstitution

Lyophilized peptide is solubilized by adding bacteriostatic or sterile water slowly down the vial wall and swirling gently — never shaking. Lyophilized material is stored at −20 °C protected from light; reconstituted material is held at 2–8 °C for short-term laboratory use, with single-use aliquots to limit freeze–thaw degradation. Research-grade reference material should carry a batch-specific Certificate of Analysis: reverse-phase HPLC for chromatographic purity (commonly ≥98%) with a chromatogram, mass spectrometry confirming molecular identity, and the name of the independent testing laboratory.
