CJC-1295 — GHRH-receptor arm

CJC-1295 is a synthetic analog of growth-hormone-releasing hormone (GHRH). It agonizes the GHRH receptor, a class B GPCR coupling to Gαs and elevating cAMP/PKA in somatotroph model systems. Variants with a Drug Affinity Complex (DAC) moiety bind albumin to extend molecular half-life in vitro, while “no-DAC” (modified GRF 1-29) forms are shorter-acting research tools.

Ipamorelin — GHSR arm

Ipamorelin is a selective pentapeptide agonist of the growth-hormone secretagogue receptor (GHSR1a), a class A GPCR coupling to Gαq/PLC. It is valued in research for its selectivity relative to earlier secretagogues.

Convergent signaling

The research rationale for the pairing is that GHRH-R (Gαs/cAMP) and GHSR1a (Gαq/Ca2+) act through distinct but convergent pathways in somatotroph models; studying them together allows characterization of additive receptor signaling within the somatotropic axis in vitro.
